Extended-spectrum cephalosporinases in Pseudomonas aeruginosa.

چکیده
The characterization of AmpC-type beta-lactamases was performed in a collection of 32 clinical Pseudomonas aeruginosa isolates with intermediate susceptibility or resistance to imipenem and ceftazidime. Twenty-one out of those 32 isolates overexpressed AmpC beta-lactamase, and the MICs of ceftazidime and imipenem were reduced after cloxacillin addition. Cloning and sequencing identified 10 AmpC beta-lactamase variants. Reduced susceptibility to imipenem, ceftazidime, and cefepime was observed only with recombinant P. aeruginosa strains expressing an AmpC beta-lactamase that had an alanine residue at position 105. The catalytic efficiencies (k(cat)/K(m)) of the AmpC variants possessing this residue were increased against oxyiminocephalosporins and imipenem. In addition, we show here that those AmpC variants constitute a favorable background for the in vitro selection of imipenem-resistant strains. This report identified a novel resistance mechanism that may contribute to imipenem resistance in P. aeruginosa.

Detection of Ampc and Extended-Spectrum Beta-Lactamases in Clinical Isolates of Pseudomonas Aeruginosa from Patients with Cystic Fibrosis
ABSTRACT Background and Objectives: Pseudomonas aeruginosa is the most frequent opportunistic pathogen isolated from the sputum of patients with cystic fibrosis (CF). Resistance to β -lactam antibiotics may arise from over expression of the naturally occurring AmpC cephalosporinases or acquired extended-spectrum β-lactamases (ESBL). The aim of...
متن کاملInhibitory Effect of Iron Oxide Nanoparticles on CTX-M Gene Expression in Extended-Spectrum βeta-Lactamase-Producing Pseudomonas aeruginosa Isolated from Burn Patients
Background and Objective: Burn wound infections caused by Pseudomonas aeruginosa exhibiting β-lactam antibiotic resistance are one of the greatest challenges of antimicrobial treatment. In this context, P. aeruginosa strains harboring resistance mechanisms, such as production of extended-spectrum beta-lactamases have the highest clinical impact no the management of burn wound infections. The ai...

Objective(s) The aim of this study was to investigate the presence of PER-1-type ESBLs in drug resistant Pseudomonas aeruginosa isolates. Materials and Methods During one-year period (2008-2009), following isolation and identification of 56 P. aeruginosa, the E-test method was performed for determination of minimal inhibitory concentration of ceftazidim. The isolates that they had MIC≥16 µg/ml ...

Objective(s) This study was carried out to evaluate the existence of the TEM, SHV and PER ESBL genes in ESBL producing strains of Pseudomonas aeruginosa isolated from burnt patients at Shafa-hospital, Kerman, Iran. Materials and Methods A total of 120 strains of P. aeruginosa were isolated from 245 patients in burn unit of Shafa-hospital during January 2006 to December 2007. MIC of antibioti...
متن کاملEvaluation of the β-Lactamase Disk Test Method in the Detection of Extended-Spectrum-β-Lactamases in Clinical Isolates of Multidrug-Resistant Pseudomonas aeruginosa
Background & Aims: The production of extended-spectrum-β-lactamases (ESBLs) is the main mechanism of resistance to β-lactam antibiotics. The outbreak of isolates simultaneously possessing several resistance mechanisms to β-lactam antibiotics caused a decrease in sensitivity of the confirmatory tests for ESBL. The aim of this study was the evaluation of the β-lactamase disk test...
